Hi all,
I need for my work in plant biology research, to take macro photographies of fungi growing on leaves. I have a Nikon D90 and a macro lens AF-S Micro NIKKOR 60 mm 1:2.8.
I would like to do focus stacking pictures since it would allow to see little fungi growing at all depth, since leaves are not flat.

https://www.youtube.com/watch?v=VGOHfDTLneg "Focus stacking with digiCamControl", which shows very good results, but the first step is the set the auto focus. When I try on my side, I click on it, but nothing happens.
It's very hard to adjust manually the focus since the layer where I can see fungi is actually very thin, so I think I really need this auto focus to work.
Did I do something wrong? The camera is on AUTO mode, while connected to the computer.
I am sorry I am very bad with cameras, I know nothing (don't judge me please, I am a geneticist )
